Quantitative Western Blots =Smart Protein Layers
The first standard-based technology for quantitative Western Blots and absolute quantification of 1D gels
.
The problem
Quantitative Western blot analysis require an appropriate reference. Recent publications show that house-keeping proteins (ß-actin, ß-tubulin, GAPDH) are regulated under various conditions.Therefore, total protein is a more accurate reference. However, as protein is washed away from the membrane during WB procedure target and reference proteins must be detected at the same point of time.

The solution
Smart Protein Layers (SPL) is the new technology for stain-free, quantitative and standardized analysis of protein gels and Western blots. For the first time, the target protein and – as a reference – the amount of total protein of this sample can be detected at the same time. In addition, SPL allows to precisely compare data derived from different experiments.

SPL combines
- the Smart Label for high sensitivity total protein visualization during every step of electrophoresis and Western blot procedure.
- the Smartalyzer, a sample-specific fluorescent standard for precise normalization and quantification of protein gels and Western blots within one experiment
- the Calibrator, a gel- and blot-specific standard to compare data from different experiments. Works also as a fluorescent MW ladder.

For 1D gels the Smart Protein Layers allow you to run as little as 1-2 µg of total protein per lane and to detect (without staining and destaining) as many protein as with the 10 fold amount of protein visualized by Coomassie staining. Compared to silver, the dynamic range (range between non-saturated and saturation) is 1,000 fold higher.
For quantitative Western blots, the Smart Protein Layers are the reliable reference for your target protein expression. To detect your target protein keep your method of choice such as chemiluminescence or the usage of primary/ secondary fluorescent antibody conjugates.
