To avoid interference of fluorescence light emission of the dyes bound to the proteins, the G-Dyes are not matched with respect to their electrophoretic mobility. Bioinformatic processing of 2D images allows both the qualitative and quantitative detection of differentially expressed proteins and of post-translational modified proteins, respectively. Therefore, for optimal analysis of multiplex-fluorescence 2D gel electrophoresis, we recommend software suitable for horizontal spot registration (warping) like Delta2D (Decodon GmbH). |

quantification of protein expression
- 2 samples labeled with fluorescent dyes (e.g. G-Dye 200, G-Dye 300)
- both samples labeled with internal standard (e.g. G-Dye 100)
- analyze protein expression by using appropriate 2D software (e.g. Delta2D/ Decodon, SameSpots/ Nonlinear Dynamics, Proteomweaver/ Bio-Rad).
